I'm not sure what you're using, but I've had good luck with the rapid ligase kit from Roche. Of course, after I left the lab, a friend figured out a DIY recipe for making your own rapid ligase kit. It's just standard T4 DNA ligase with a different buffer. (Recipe is on BenchFly- justsearch "rapid ligase" if you're interested)<br /><br />Best of luck completing the project and solving the image problem!
